Response of Human Myoblasts to Therapeutic
نویسنده
چکیده
Objectives: The aim of this study was to investigate the response of human skeletal muscle myoblasts to therapeutic doses of X-irradiation. Methods: Myoblasts was established via skeletal muscle biopsies which irradiated in vitro by x-ray at doses 3, 5 and 7 Gy. Two effects of radiation were measured (acute effect after 24h and long term effect after 72h). Acute effect was monitored by interleukin-6 release, while long term effect was monitored by measuring the proliferation capacity and cell death. Results: When comparing the non-irradiated control and cells processed with inhibitor of cell proliferation Ara C, myoblast proliferation reduced in 72h post-radiation, this change was significant with increasing doses. Post-irradiation myoblast survival determined by measurement of released LDH enzyme activity showed increased activity after exposure to x-rays. The acute reaction of myoblasts to lower doses of irradiation (3 and 5 Gy.) was reduced secretion of constitutive interleukin-6. Conclusion: The current study approved that there was positive response of human skeletal muscle myoblasts to therapeutic doses of X-irradiation..

Therapeutic effect of psoralen on muscle atrophy induced by tumor necrosis factor-α
Objective(s): To observe and determine the effect and mechanism of psoralen on tumor necrosis factor-α (TNF-α)-induced muscle atrophy.Materials and Methods: Three sets of C2C12 cells, including blank control, TNF-α (10 or 20 ng/ml) treatment and a TNF-α (10 or 20 ng/ml) plus psoralen (80 μM) administration were investigated. Cell viabili...
متن کاملIsolation of human foetal myoblasts and its application for microencapsulation
Foetal cells secrete more growth factors, generate less immune response, grow and proliferate better than adult cells. These characteristics make them desirable for recombinant modification and use in microencapsulated cellular gene therapeutics. We have established a system in vitro to obtain a pure population of primary human foetal myoblasts under several rounds of selection with non-collage...
متن کاملEffect of ionizing radiation on human skeletal muscle precursor cells
BACKGROUND Long term effects of different doses of ionizing radiation on human skeletal muscle myoblast proliferation, cytokine signalling and stress response capacity were studied in primary cell cultures. MATERIALS AND METHODS Human skeletal muscle myoblasts obtained from muscle biopsies were cultured and irradiated with a Darpac 2000 X-ray unit at doses of 4, 6 and 8 Gy. Acute effects of r...
متن کاملTherapeutic Response to Folinic Acid in Me-thanol Poisoning Epidemic in Shiraz
Background: Methanol poisoning may result to death and permanent complications, sporadic cases of methanol poisoning are fairly rare and the largest experience are generally gained from managing methanol intoxication epidemics. The main metabolite of methanol is acid formic that is the prime metabolite responsible for toxic effects of methanol and since folate dependent systems are responsible ...
متن کاملTransient immunosuppressive treatment leads to long-term retention of allogeneic myoblasts in hybrid myofibers
Normal and genetically engineered skeletal muscle cells (myoblasts) show promise as drug delivery vehicles and as therapeutic agents for treating muscle degeneration in muscular dystrophies. A limitation is the immune response of the host to the transplanted cells. Allogeneic myoblasts are rapidly rejected unless immunosuppressants are administered. However, continuous immunosuppression is asso...
